How Does Emgality Block CGRP?

Galcanezumab-gnlm binds to calcitonin gene-related peptide, commonly called CGRP, and blocks the ligand from binding to its receptor. CGRP signaling is involved in migraine and cluster headache biology. Blocking this pathway reduces CGRP receptor activation.

Who Is Treated With Emgality?

Emgality is approved for adults who need preventive migraine treatment and adults with episodic cluster headache. Current U.S. labeling does not establish safety and efficacy in pediatric patients. Migraine and cluster headache use different dose strengths and injection schedules.

How Is Emgality Dosed?

For migraine prevention, treatment starts with 240 mg as two consecutive 120 mg injections, followed by 120 mg once monthly. For episodic cluster headache, 300 mg is given as three consecutive 100 mg injections at the start of the cluster period and then monthly until the cluster period ends.

What Serious Risks Are Linked to Emgality?

Serious hypersensitivity reactions, including anaphylaxis and angioedema, have been reported. Current labeling also warns about constipation with serious complications, new or worsening hypertension, and new or worsening Raynaud phenomenon. Injection-site reactions are the most common reactions reported in clinical studies.
