How Does Desloratadine Reduce Allergy Symptoms?

Desloratadine is a long-acting tricyclic antihistamine with selective activity at peripheral H1 receptors. Blocking histamine at these receptors reduces symptoms such as sneezing, runny nose, nasal itching, watery eyes, itching, and hives. Desloratadine also inhibits histamine release from human mast cells in laboratory studies.

Which Conditions Are Treated With Desloratadine?

Desloratadine treats seasonal and perennial allergic rhinitis and chronic idiopathic urticaria. The 5 mg tablet is labeled from age 12. Current oral liquid labeling covers seasonal allergic rhinitis from age 2 and perennial allergic rhinitis and chronic idiopathic urticaria from age 6 months.

How Is Desloratadine Dosed?

Adults and adolescents age 12 and older take 5 mg once daily with or without food. Oral liquid dosing is 2.5 mg once daily for ages 6 to 11, 1.25 mg once daily for ages 1 to 5, and 1 mg once daily for ages 6 to 11 months. Adults with liver or kidney impairment start with 5 mg every other day.

What Safety Problems Need Attention With Desloratadine?

Serious hypersensitivity reactions, including rash, itching, hives, swelling, breathing difficulty, and anaphylaxis, have been reported. Desloratadine is contraindicated in patients with hypersensitivity to desloratadine, loratadine, or product ingredients. Other reported reactions include dry mouth, fatigue, muscle pain, throat inflammation, and sleepiness.
