How Does Dayvigo Promote Sleep?

Lemborexant blocks orexin receptors OX1R and OX2R. Orexin A and orexin B are neuropeptides involved in promoting wakefulness. Blocking their binding suppresses wake-related signaling, which helps patients fall asleep and stay asleep. The labeled mechanism differs from medicines that act mainly through GABA receptors.

Who Is Treated With Dayvigo?

Dayvigo is used in adults with insomnia involving sleep-onset difficulty, sleep-maintenance difficulty, or both. It is contraindicated in patients with narcolepsy. The effect on breathing is considered in patients with impaired respiratory function, including obstructive sleep apnea or chronic obstructive pulmonary disease.

How Are Dayvigo Tablets Dosed?

The recommended dose is 5 mg no more than once per night immediately before bedtime, with at least seven hours remaining before planned awakening. The dose can increase to 10 mg according to response and tolerability. Taking Dayvigo with or soon after food can delay the time to sleep onset.

What Serious Risks Are Linked to Dayvigo?

Dayvigo can impair alertness and motor coordination the next day, especially at the 10 mg dose. Other concerns include sleep paralysis, hallucinations around sleep transitions, cataplexy-like symptoms, complex sleep behaviors, worsening depression, suicidal thinking, and additive sedation with alcohol or other central nervous system depressants.
