How Does Cinryze Help Prevent HAE Attacks?

C1 inhibitor is a normal blood protein that regulates complement, contact-system, and fibrinolytic pathways. People with hereditary angioedema have low levels or reduced function of this protein. Cinryze raises functional C1 inhibitor activity and suppresses contact-system activation, which reduces bradykinin generation and vascular permeability linked to swelling attacks.

Who Is Treated With Cinryze?

Cinryze is approved for routine prophylaxis against hereditary angioedema attacks in adults, adolescents, and children age 6 and older. It is used to reduce attack frequency rather than to treat an acute attack after symptoms have started. Pediatric dosing differs from dosing for patients age 12 and older.

How Is Cinryze Dosed?

Adults and adolescents age 12 and older receive 1000 IU intravenously every three or four days. Children age 6 to 11 receive 500 IU every three or four days. Dose increases are permitted for inadequate response within current labeling limits. Infusion time is about 10 minutes for 1000 IU and 5 minutes for 500 IU.

What Serious Risks Are Linked to Cinryze?

Serious hypersensitivity reactions, including anaphylaxis, can occur during or after infusion. Arterial and venous thromboembolic events have also been reported, especially in patients with underlying risk factors. Since Cinryze is made from human plasma, a residual risk of transmitting infectious agents remains despite donor screening and viral reduction steps.
