How Does Camzyos Affect Cardiac Myosin?

Mavacamten reversibly inhibits cardiac myosin by binding to myosin and reducing the number of myosin heads available to interact with actin. This decreases excessive actin-myosin cross-bridge formation and reduces excessive heart muscle contraction associated with obstructive hypertrophic cardiomyopathy.

Who Is Treated With Camzyos?

Camzyos is approved for adults with symptomatic New York Heart Association class II or III obstructive hypertrophic cardiomyopathy. Treatment is individualized according to left ventricular ejection fraction, symptoms, and echocardiographic measurements. Starting treatment is not recommended when left ventricular ejection fraction is below 55 percent.

How Is Camzyos Dosed and Monitored?

Camzyos is taken once daily, with dose selection based on clinical status and echocardiographic findings. Available strengths are 2.5 mg, 5 mg, 10 mg, and 15 mg. Heart function is assessed before and during treatment. Therapy is interrupted if left ventricular ejection fraction falls below 50 percent or clinical status worsens.

What Serious Risk Is Linked to Camzyos?

Camzyos carries a boxed warning for heart failure caused by systolic dysfunction. Drug interactions that increase mavacamten exposure can raise this risk, while enzyme inducers can reduce treatment activity. Echocardiographic monitoring is required before and during therapy. Certain CYP2C19 and CYP3A4 inhibitors or inducers are contraindicated with Camzyos.
