What Are Glycoprotein IIb/IIIa Inhibitors Used For?

Glycoprotein IIb/IIIa inhibitors are used in selected patients with acute coronary syndromes or during percutaneous coronary intervention. They can reduce thrombotic complications when rapid and potent platelet inhibition is needed. Current use is commonly limited to specific high-risk or procedural situations because other antiplatelet strategies are available. The decision depends on bleeding risk, kidney function, prior medicines, and findings during the procedure.

How Do Glycoprotein IIb/IIIa Inhibitors Work?

Platelet activation changes the glycoprotein IIb/IIIa receptor into a form that can bind fibrinogen. Fibrinogen then bridges neighboring platelets and helps build a platelet plug. Glycoprotein IIb/IIIa inhibitors occupy or block this receptor and prevent the bridging step. Their effect begins rapidly when the medicine is given intravenously.

How Are Glycoprotein IIb/IIIa Inhibitors Given?

These medicines are administered through a vein, commonly as an initial dose followed by an infusion. Dosing can depend on body weight, kidney function, and the clinical procedure. They are frequently used with aspirin, an anticoagulant, or another antiplatelet medicine under close supervision. Platelet counts, hemoglobin, bleeding, vascular access sites, and kidney function can require monitoring.

What Are the Side Effects and Precautions?

Bleeding is the main adverse effect and can occur at an access site, internally, or in another location. A rapid fall in platelet count can also occur and can be severe. Risk increases with other antithrombotic medicines, invasive procedures, older age, kidney impairment, or a history of bleeding. Active internal bleeding and certain recent bleeding events can make treatment inappropriate.
