How Does Entyvio Affect Gut Inflammation?

Vedolizumab binds the alpha4beta7 integrin on selected memory T lymphocytes and blocks its interaction with MAdCAM-1 on gut endothelial cells. This inhibits migration of these lymphocytes into inflamed gastrointestinal tissue and reduces a pathway involved in chronic intestinal inflammation.

Who Is Treated With Entyvio?

Entyvio treats adults with moderately to severely active ulcerative colitis or Crohn disease. Before treatment, tuberculosis assessment is considered and immunizations are updated. Current U.S. labeling does not establish pediatric safety and efficacy.

How Is Entyvio Dosed?

Treatment starts with 300 mg by intravenous infusion at week 0 and week 2. At week 6, patients can receive another 300 mg intravenous infusion followed by 300 mg every eight weeks, or switch to 108 mg subcutaneously every two weeks. Treatment is discontinued when there is no therapeutic benefit by week 14.

What Serious Risks Are Linked to Entyvio?

Serious concerns include infusion-related or hypersensitivity reactions, infections, liver injury, and a potential risk of progressive multifocal leukoencephalopathy. Treatment does not start during a clinically significant active infection. A serious infection that develops during therapy calls for withholding Entyvio until the infection resolves.
