What Is Dupixent?

Dupixent is a prescription monoclonal antibody containing dupilumab. It is an interleukin-4 receptor alpha antagonist given by subcutaneous injection. Current U.S. indications span several type 2 inflammatory diseases, including atopic dermatitis, asthma, chronic rhinosinusitis with nasal polyps, eosinophilic esophagitis, prurigo nodularis, COPD, chronic spontaneous urticaria, bullous pemphigoid, and allergic fungal rhinosinusitis.

How Does Dupixent Block Type 2 Inflammation?

Dupilumab binds the interleukin-4 receptor alpha subunit shared by the IL-4 and IL-13 receptor complexes. This blocks IL-4 signaling and IL-13 signaling through the relevant receptor pathways. Reduced signaling in these pathways lowers type 2 inflammatory activity involved in several labeled diseases.

Who Is Treated With Dupixent?

Current labeling includes atopic dermatitis from age 6 months, asthma from age 6, eosinophilic esophagitis from age 1 in patients weighing at least 15 kg, chronic spontaneous urticaria from age 2, and allergic fungal rhinosinusitis from age 6 after sinonasal surgery. Other indications have their own age and disease criteria.

How Is Dupixent Dosed?

Dosing depends on the condition, age, and body weight. Adults with atopic dermatitis receive a 600 mg starting dose followed by 300 mg every two weeks. Other indications use schedules ranging from every week to every four weeks, with weight-based regimens for several pediatric uses.

What Serious Risks Are Linked to Dupixent?

Serious concerns include hypersensitivity reactions, conjunctivitis, keratitis, blepharitis, eosinophilic conditions, new-onset psoriasis, joint symptoms, and parasitic infections. Dupixent does not treat acute asthma or COPD symptoms. Live vaccines are avoided immediately before and during therapy.
