How Does Colazal Act in the Colon?

Balsalazide reaches the colon largely intact, where intestinal bacteria split it into mesalamine and an inert carrier. Mesalamine acts locally on the colonic lining and reduces inflammatory activity. Its exact anti-inflammatory mechanism is not fully defined.

Who Is Treated With Colazal?

Colazal treats mildly to moderately active ulcerative colitis in patients age 5 and older. Safety and efficacy beyond eight weeks have not been established in patients age 5 to 17. In adults, current labeling states that safety and efficacy beyond 12 weeks have not been established.

How Are Colazal Capsules Dosed?

Adults take three 750 mg capsules three times daily for up to eight weeks, with some clinical-trial patients treated for up to 12 weeks. Patients age 5 to 17 use either three capsules three times daily or one capsule three times daily for up to eight weeks. Capsules can be swallowed whole or opened onto applesauce.

What Safety Problems Need Attention With Colazal?

Serious concerns include kidney impairment, mesalamine-induced acute intolerance syndrome, hypersensitivity reactions, liver failure, severe skin reactions, upper gastrointestinal obstruction, photosensitivity, and kidney stones. Kidney function is checked before treatment and periodically during therapy. Adequate fluid intake reduces the risk of kidney stone formation.
