How Does Aspirin Affect Pain and Platelets?

Aspirin inhibits cyclooxygenase enzymes and reduces production of prostaglandins involved in pain, fever, and inflammation. It also irreversibly inhibits platelet cyclooxygenase, reducing formation of thromboxane A2, a substance involved in platelet aggregation and blood vessel constriction. The platelet effect lasts for the life of the affected platelet.

When Is Aspirin Used?

Regular-strength 325 mg aspirin is used for temporary relief of headache, muscle pain, minor arthritis pain, toothache, menstrual pain, and pain or fever from colds. Low-dose aspirin is also taken under clinician direction for selected antiplatelet uses. Delayed-release 81 mg products act too slowly for rapid relief of headaches or similar symptoms.

How Are Regular-Strength Aspirin Tablets Taken?

For a 325 mg over-the-counter tablet, adults and children age 12 and older take one or two tablets every four hours or three tablets every six hours. The label limits use to 12 tablets in 24 hours unless a doctor directs otherwise. Each dose is taken with a full glass of water.

What Safety Problems Need Attention With Aspirin?

Aspirin can cause severe stomach bleeding and serious allergic reactions. Children and teenagers recovering from chickenpox or flu-like illness should not use aspirin due to Reye syndrome risk. Ringing in the ears can signal toxicity. Pregnancy, kidney disease, liver disease, asthma, bleeding disorders, anticoagulant use, and other NSAID use call for medical review before treatment.
