How Does Apriso Act in the Colon?

The exact mechanism of mesalamine is not fully known. Its action appears to be a local anti-inflammatory effect on cells lining the colon. Mesalamine is thought to reduce inflammation partly by decreasing production of arachidonic acid metabolites, including prostanoids and leukotrienes, that are elevated in ulcerative colitis.

Who Uses Apriso for Ulcerative Colitis?

Apriso is approved for adults who need maintenance treatment to keep ulcerative colitis in remission. Its U.S. indication is maintenance of remission rather than induction treatment for active ulcerative colitis. Kidney function is checked before treatment and periodically during therapy due the risk of renal injury.

How Should Apriso Capsules Be Taken?

The recommended adult dose is four 0.375 g capsules once daily in the morning, for a total of 1.5 g. Capsules are swallowed whole and should not be cut, broken, crushed, or chewed. Apriso can be taken without regard to meals. Antacids are avoided, and patients are advised to drink adequate fluids.

What Safety Problems Need Attention With Apriso?

Risks include kidney impairment, mesalamine-induced acute intolerance syndrome, hypersensitivity reactions, liver failure in people with liver disease, severe skin reactions, photosensitivity, and kidney stones. Each capsule contains phenylalanine from aspartame, which matters for people with phenylketonuria. New abdominal pain, bloody diarrhea, rash, fever, or reduced kidney function calls for medical review.
