How Does Agamree Work?

Vamorolone acts through the glucocorticoid receptor and produces anti-inflammatory and immunosuppressive effects. The exact mechanism responsible for its clinical effect in Duchenne muscular dystrophy is not known. Corticosteroid activity can also suppress the hypothalamic-pituitary-adrenal axis, which is one reason treatment lasting longer than one week is tapered rather than stopped abruptly.

What Is Agamree Used For?

Agamree is approved for Duchenne muscular dystrophy in patients age 2 and older. Duchenne muscular dystrophy is a genetic muscle disorder marked by progressive muscle weakness. Agamree is a corticosteroid treatment used as part of medical management for the disease. The labeled indication applies to pediatric and adult patients who meet the age requirement.

How Is Agamree Taken?

Agamree is taken by mouth once daily, preferably with a meal. The recommended dose is 6 mg per kg of body weight, capped at 300 mg daily for patients weighing more than 50 kg. Lower doses are used for mild to moderate liver impairment or with strong CYP3A4 inhibitors. The bottle is shaken well before each measured dose.

What Are the Side Effects and Warnings?

Agamree can suppress adrenal function and raise infection risk. Other concerns include increased blood sugar, elevated blood pressure, sodium or potassium changes, gastrointestinal perforation, behavioral or mood changes, decreased bone mineral density, cataracts, glaucoma, altered growth in children, blood clots, and anaphylaxis. Live vaccines are avoided during treatment. Abrupt withdrawal after longer treatment can trigger adrenal insufficiency.
